Genetic diversity of four large yellow croaker(Pseudosciaena Crocea Richardson) populations using microsatellite markers

In present study,15 microsatellite markers were used to analyze the genetic diversity of four cultured stocks(Daiqu,Min-Yue stocks and two groups of their hybrids) of large yellow croaker(Pseudosciaena Crocea Richardson).The results showed that all microsatellite markers were polymorphic in four populations.A total of 206 alleles were detected in 171 individuals,and the allele number was ranged from 7 to 23 at 15 loci.The average allele number was 13.73 with the allele sizes of 88 to 318 bp.The value of average observed heterozygosity(Ho) in four populations was 0.7319 to 0.7944,the value of average expected heterozygosity(He) was 0.5981 to 0.6893,and the value of polymorphism information content(PIC) was 0.7138 to 0.7836.The statistics indicated that all tested loci were of a plenty of polymorphisms and abundant genetic diversity in each population.The cluster analysis by UPGMA method showed that Min-Yue stock and MD group were of a close relationship.This analysis of genetic diversity using microsatellite markers could be very useful in studying species conservation for large yellow croaker.
