The invention discloses a detection method of a predictive marker Capn4 of postoperative recurrence of liver cancer. The method comprises the following steps: placing a capn4 antibody into a reaction hole of a kit and embedding overnight; adding patient samples in four holes and adding standard albumen supernatant samples in four holes; adding coating buffer liquor; mixing for 1 hour; adding a pan-capn4 antibody and a non-specific antigen; after incubation and washing, adding a horse radish peroxidase labeled antibody; then, adding a substrate; after termination of the reaction, reading the OD (Outside Diameter) value of each reaction hole in the experiment; drawing a standard curve by using measurement values of the standard samples; reading corresponding numerical values; and working out the average value, wherein the detection result shows positive if the numerical values of the patient samples exceed 1.5 times the numerical values of the standard samples. By adopting the method, high throughput and promptness of detecting individuals are taken into consideration, and quantitative detection is realized, so that the subjective error can be reduced because of mechanical detection and value reading. The invention further discloses the kit based on the method. The kit comprises 96 reaction holes arranged into a matrix, and the capn4 antibody is placed in each reaction hole.
