Genomic DNA extraction of Calanus sinicusin different preser-vations

Extraction of genomic DNA from preserved specimen is a key step to study phylogeny and molecular ge-netics of zooplankton.This paper compared the effect of different preservations on extraction of genomic DNA adopted a small crustacean copepod,Calanus sinicus,collected from Xiamen waters.Except the specimen fixed in5%of formalin solution,genomic DNA was well extracted by using the specimen fixed in95%ethanol and placed at4℃,100%ethanol at4℃and TE solution at-80℃.The genomic DNA can be used as a template for PCR process of mtCOI gene fragment.
