The effect of PMMA particles on the expression of RANK in rats macrophage

Objective To observe the effect of PMMA particles on the expression of RANK in rats macrophage cultured in vitro. Methods Peritoneal macrophages were obtained from the peritoneal of rats and cultured and purified in vitro.Cells purity were identified by immunocytochemistry techniques and treated with PMMA particles according to the experimental design. Semi-quantitative reverse transcription PCR method was used to detect the level of RANK mRNA. Results PMMA particles enhanced RANK mRNA expression of rats peritoneal macrophage in a time-and dose-dependent fashion.RANK mRNA expression in the peritoneal macrophage was elevated dose-dependently and time-dependently following PMMA particles treatment and reached its peak level at a concentration of 0.1‰～5‰(v/v)(P0.05),especially co-cultured for 24 h in vitro (P0.05). Conclusion By altering the relative expression of RANK mRNA,PMMA particles have the potential of facilitating macrophage transform into cells that acting as bone resorption ones.
