Effects of small interfering RNA targeting Dicer enzyme on the biological behavior of human glioma cells

Objective To observe the effects of knocking down Dicer enzyme expression on the biological behaviors of human glioma cells U87 and U251.Methods Using small interference RNA (siRNA) technology,Dicer enzyme expression was knocked down,Western blot detection U87 and U251 cells' Dicer enzyme expression after transfected with small interfering RNA 24 h; application of cell counting kit-8 (CCK-8) assay,Tranwell test and gelatinase spectrum experiments to detect cell proliferation,invasion and matrix metalloproteinase [matrix metalloproteinase (MMP)-2,MMP-9] activity.Results Transfected with siRNA targeting Dicer enzyme,Dicer enzyme compared with the control group decreased by 68.0% (U87) and 60.3% (P <0.05) (U251).Both cell proliferation rate was significantly higher than the corresponding control group,and with time gap also increased (P < 0.05).number of cell invasion siRNA group 77.9±8.3 (U87) and 81.0 ±7.9 (U251) compared with negative control group 40.6 ±7.0 (U87) and 35.8 ± 6.5 (U251) increased significantly (P < 0.05).The siRNA set of MMP-2 activity increased compared with the control group (1.35 ± 0.17) times (U87),(1.19 ± 0.06) times (U251) (P < 0.05) ; MMP-9 activity increased compared with the control group (1.72 ± 0.37) times (U87),(1.66 ± 0.33) times (U251) (P < 0.05).Conclusion After dicer enzyme expression knockdown,human glioma cell proliferation increased,MMP-2,MMP-9 activity was enhanced and invasive increased.
