Isolation of an anti-tumor compound from the leaves of Annnona muricata and pharmacological characterization in PC-3 cells

Annona muricata L (Annonaceae), commonly known as soursop, has a long history in herbal medicine with a lengthy recorded indigenous use. It has also been found to be a promising new anti-tumor agent in numerous in vitro studies. The present investigation concerns the antiproliferative effects of one isolated active compound from the leaves of Annona muricata in human PC-3 cancer cells. One of the active compounds was isolated from the crude extract of Annona muricata leaves by using a cell viability-guided chromatography technique. The Isolated compound was characterized by C-NMR and H-NMR and was found to be “Rolledecin C’ which was previously reported by Zheng-Ming Gu et al. PC-3 cells seeded in 96 well plates were treated with different concentrations from 1 to 100 μg/ml of the isolated compound. Cell viability was measured after 24 hours of incubation showing that the isolated compound reduced cell viability in a dose-dependent manner with an IC50 lower than 10 μg/ml.
