The invention discloses a detection kit and a method for non-structural protein antibody dot blots of foot and mouth disease viruses. The detection kit comprises five NSP (Non Starch Polysaccharides) blot membrane strips, serum diluent, a 25* concentrated PBST cleaning solution, a membrane color development substrate solution, 100* concentrated enzyme-labeled detection antibodies, positive control serum, negative control serum and a serum dilution plate with 20 holes. According to the method, five NSP antibodies are simultaneously detected, the differential diagnosis accuracy of foot and mouth disease infected animals and immune animals can be further improved. The method can be used as a confirmation and diagnosis method of 3ABC antibody detection positive and suspicious animals, has the advantages of higher sensitivity and specificity and is suitable for detecting the serum of three susceptible animals of cattle, sheep and pigs.
