Tumor inhibitory and immunoloregulation effects of Tiaoheng Prescription on S180 ascites of tumor-bearing mice

2010
Objective To investigate the mechanisms of tumor Inhibitory and immunoloregulation of "Tiaoheng Prescription" on S180 ascites of tumor-bearing mice. Methods The mouse model of S180 ascites of tumor-bearing mice were established and then the mice were given Tiaoheng Formula by intragastric administration. Twelve days later, the mice were executed and the ascites smears were prepared. The cell feature in ascites was observed under light microscope, and the expression of CD4 and CD8 on the cellular membrane of the T lymphocyte in ascites and the level of IL-10 in the peripheral blood was detected with immunohistochemical or ELISA method. Results A large amount of damaged and apoptotic S180 ascites cells were found in Tiaoheng Prescription middle dosage group, and the changes of cell feature in control group were not obvious. The expressions of CD4 and CD8 on the membrane of T cells in ascites was increased in masculine group, Tiaoheng Prescription middle and large dosage groups, and CD8 expression was increased in heat-shock group. The production of IL-10 by immunocyte obviously decreased in Tiaoheng Prescription[25、50 g/(kg·d) by gavage for 12 days]-treated group(P0.05,P0.01).Conclusion Tiaoheng Formula can inhibit the cancer cell of S180 ascites in mice. The mechanism may be related to that Tiaoheng Formula can enhance T lymphocyte immunological function by regulating the expression of T lymphocyte CD4 and CD8 molecules in S180 ascites, inhibit the activity level of IL-10 in the peripheral blood, and to prohibit the invasion and metastasis of tumor cells.
