The detection of TetM of N.gonorrhoeae and analysis of the plasmid intermediate drug resistance

Objective To set up a PCR method to detect the TRNG strain of N. gonorrhoeae and investigate the plasmid intermediate drug resistance of N. gonorrhoeae. Methods Detection of the TRNG strain of N. gonorrhoeae by design distinctive primer. The MIC of penicillin and tetracycline to 153 strains were determined by agar dilution. Results 83 TRNG strains by identified MIC were PCR positive, the others are negative. Using total amount of plasmid or the cell suspensions of N. gonorrhoeae to be the detection model. The lowest percentage of detection was 0.1pg or 100 N. gonorrhoeae cells, respectively. The investigation of N. gonorrhoeae's plasmid intermediate resistance showed: the percentage of plasmid intermediate resistance was 87%, with PPNG 71%, TRNG 54%, PPNG/TRNG 39%. PPNG/TRNG has been the main epidemic strains of this area. Conclusion Detection of TRNG strain by PCR is extremely special and sensitive. Comparing with traditional methods, it is convenient for clinical application, fast, accurate, and reliable clinical. The plasmid intermediate drug resistance of N. gonorrhoeae appears high tendency in Chengdu.
