Determination of amino acids in Koelrenteria paniculata fruit and Koelreuteria integrifoliola Merr fruit by reversed-phase high performance liquid chromatography with precolumn derivatization

In order to determine the composition and contents of amino acids in Koelrenteria paniculata fruit and Koelreuteria integrifoliola Merr fruit from zhengzhou by reversed-phase high performance liquid chromatographic (RP-HPLC ) method ,an Ultimate Amino Acid column was used ,and phenylisothiocyanate (PITC ) was the precolumn derivatization agent. Elutions were carried out in a binary gradient mode. The Column temperature was 30℃ ,and detection wavelength was set at 254nm. Results showed that:Koelrenteria paniculata fruit and the Koelreuteria integrifoliola Merr fruit contained 17 kinds of amino acids,total amino acid content was 15.57% and 17.53% respectively,The recoveries of standard addition of amino acids were 80.5%~115.7%. The average recovery rate was 93.86% ,glutanic acid content was the highest. In Koelrenteria paniculata fruit,the histidien content was the lowest,the isoleucine content in Koelreuteria integrifoliola Merr fruit was the lowest. This method was simple,accurate and efficient. Measurement results were satisfactory,which provided reference for the development and utilization of the wild resources.
