Inhibition effects of adenovirus vector-mediated RNA interference on expression of AFP gene in FU97 gastric carcinoma cells

OBJECTIVE:To investigate the inhibition of AFP gene in gastric carcinoma (AFP-GC) cell FU97 by recombinant adenovirus vector of siRNA specific for Alpha-fetoprotein,and study the influence to the tumor cell on the growth cycle and apoptosis after silencing the AFP gene.METHODS:The Ad-EGFP-U6-AFP-siRNA was transfected into FU97.An optimized mutiplicity of infection was chosen.RESULTS:The optimized MOI was selected(MOI=100).AFP in the cells was obviously decreased on mRNA and protein level after being transfected with Ad-EGFP-U6-AFP-siRNA;The cells in G0/G1 phase〔(33.49±1.85)%〕 were notably fewer than that in the negative control and control〔(64.76±2.98)% and (74.81±2.60)%,P0.01〕.And the cells in G2/S phase of RNAi group〔(64.10±2.59)%〕notably higher than that in the negative control and control 〔(34.77±1.91)% and (29.85±2.85)%,P0.01〕.But there was no difference between RNAi group and controls on apoptosis percentage(P0.05).CONCLUSIONS:RNA interference mediated by recombinant adenovirus can effectively inhibit AFP expression in FU97 cells on mRNA and protein level.After silencing the AFP gene,cell generation cycle is affected.But there is no obviously affected on apoptosis.
