Anti-idiotypic antibodies induce neutralizing antibodies to bovine herpesvirus 1.

SUMMARY A neutralizing murinemonoclonal antibody (mAb)oftheIgG2aisotype (MM-113),specific for bovine herpesvirus I(BHV-1)glycoprotein gIV, was usedtodevelop anti-idiotypic antibodies (antiId)ina calf. Thebovine anti-Id were isolated fromtheserum oftheimmunized calf byaffinity chromatography on an MM-I13-Sepharose column, followed byrepeated adsorption on a murine IgG2acolumn. Theanti-Id thusobtained specifically reacted withMM-113,butnotwithisotypematchedcontrols. Theyalsoinhibited thebinding ofMM-113toBHV-1in a concentrationdependent manner. Miceimmunized withtheanti-Id produced neutralizing antibodies toBHV-1. Theanti-Id boundtocells permissive toBHV-1 ina cell-binding radioimmunoassay (RIA).
