During cancer development and progression, tumor cells undergo abnormal epigenetic modifications, including DNA methylation and histone deacetylation. These aberrations promote genomic instability and lead to silencing of tumor-suppressor genes; methylation status of some genes serves as the diagnostic marker for cancer. Somatic hypermethylation of CpG island sequences at GSTP1, the gene encoding the pi-class glutathione S-transferase, appears to be characteristic of human prostatic carcinogenesis. We present here a review of the literature describing the role of epigenetic alteration in cancer development and somatic GSTP1 changes in DNA from prostate cells and tissues.
