Flow fluorescent coding microballon-based avian influenza virus multi-type simultaneous detection method and kit

2010
The invention discloses flow microballon technology-based simultaneous immunoassay method and kit for detecting avian influenza viruses in different subtypes. The simultaneous immunoassay method is a flow liquid phase immunoassay method which is used for simultaneously detecting the avian influenza viruses in different subtypes by using a polystyrene fluorescent coding microballon as a solid phase carrier and a quantum dot as a fluorescent labeling substance. The method comprises the following steps of: covalently combining a coated antibody on the surface of the coding microballon; in a filter membrane plate, specifically combining the coated antibody, viruses, a detection antibody and a secondary antibody to form a fluorescent immune compound body of microballon-coated antibody conjugate-viruses-detection antibody-quantum dot labeled secondary antibody (or microballon-coated antibody conjugate-viruses-detection antibody-quantum dot labeled detection antibody); subjecting the compound body to pass through a detection area of the flow analysis system; identifying different coding microballons and detecting the fluorescence intensity of the quantum dot; and calculating the average fluorescence intensity ratio to finish the detection. By connecting avian influenza virus monoclonal antibodies or polyclonal antibodies in different subtypes through different coding microballons, the invention can be used for simultaneously detecting various avian influenza viruses in different subtypes or simultaneously detecting the avian influenza viruses in different subtypes in various samples and has the advantages of quickness and high throughout.
