Subcellular Localization and Expression of MADS-box Gene NMH7 in Alfalfa

The subcellular location of gene NMH7 in alfalfa was studied,a recombinant plasmid with a green fluorescent protein gene(GFP-NMH7) was constructed and transformed into onion epidermis cells by gene gun method.Transient expression analysis indicated that GFP-NMH7 fusion protein was localized in the nucleus of onion epidermal cells.Semi-quantitative RT-PCR analysis was used to determine the tissue expression patterns of gene NMH7 in alfalfa,the results revealed that it expressed itself in roots,floral buds,flowers and pods,and there was the highest expression level in flowers.The results of Semi-quantitative RT-PCR analysis showed that the expression of gene NMH7 in alfalfa was not induced by gibberellin and the expression of NMH7 gene in alfalfa was inhibited by Jasmonic acid methyl ester.
