The dynamic study of hyperlipidemic fatty liver in mice

2009
Aim:To establish the hyperlipidemic fatty liver model in mice and observe the dynamic change of hepatic steatosis in different time. Methods: 50 Kunming mice were randomly divided into control group(n1=30) and high-fat diet(HF) group(n2=30).The mice were fed with standard diet and high-fat diet respectively. The serum TC,TG,HDL-C,LDL-C and hepatic TC,TG were detectd dynamically from 2 to 4 weeks. At the same time, liver index was observed and hepatic steatosis was analyzed by HE and Sudan Ⅲ staining. Results: The serum TC,HDL-C,LDL-C,TC/HDL-C and hepatic TC,TG were elevatd significantly in HF group after 2 weeks. In the following 3-4 weeks, the serum lipid, hepatic lipid and liver index kept rising.Hepatic steatosis was induced, which the degree of steatosis was aggravated with time going on.Conclusion:Feeding with high-fat diet, different phases of fatty liver had been developed in 4 weeks, which establishes favourable bases for further study on drugs of anti-fatty liver.
