EXPERIMENTAL STUDY Neuroprotective effects of Fructus Chebulae extracts on experimen- tal models of cerebral ischemia

Abstract OBJECTIVE: Toinvestigatetheneuroprotectiveef-fectsof FructusChebulae extractusingboth invivoandinvitromodelsofcerebralischemia.METHODS: Asan invitro model,oxygenglucosedeprivationfollowedbyreoxygenation(OGD-R)andhydrogenperoxide(H 2 O 2 )inducedcellulardamageinratpheochromocytoma(PC12)cellswasusedtoinvestigatetheneuroprotectiveeffectsofextractof FructusChebulae. 3-(4,5-dimethylthia-zol-2-yl)-2,5-diphenyltetrazolium bromide assaywasusedtocalculatecellsurvival.Forinvivo,occlu-sionofleftmiddlecerebralarteryonratswascar-riedoutasafocalcerebralischemicmodel.RESULTS: FructusChebulae extractincreasesthePC12 cellsurvivalagainstOGD-RandH 2 O 2 by 68%and 91.4%respectively. FructusChebulae alsode-creasesthecerebralinfarctvolumeby 39%andex-tentofhemisphereswellingfrom 17%incontrolgroupto10%inFructusChebulaetreatedgroup.CONCLUSION: FructusChebulae,asatraditionalmedicine,canrescuetheneuronalcelldeathagainstischemiarelateddamage.Thepossiblemechanismfortheneuroprotectionmightbetheinhibitionofoxidativedamagesoccurringafteracutephaseofcerebralischemia.©2014JTCM.Allrightsreserved.Keywords: FructusChebulae;Pheochromocytoma;Neuroprotection;Brainischemia;Middlecerebralartery
