Isolation and identification of bovine viral diarrhea virus.

【objective】In order to understand the prevalence status of Bovine viral diarrhea/Mucosal disease(BVD/MD) in Xinjiang,an experiment was carried out to isolate Bovine viral diarrhea virus(BVDV)from the speciment of suspicious cases.【Method】Specimens collected from the beef farms of different areas in Xinjiang were inoculated by MDBK cells and reproduced blindly for 2-4 generations;Toxicity and serum neutralization test(SNT) of isolated strains were conducted,and 5'UTR genes were amplified by reverse transcription-polymerase chain reaction(RT-PCR) and analyzed after cloning and sequencing.【Result】Eight strains of them could cause cells pathological changes with the TCID50 between 103 and 106.The result of serum neutralization test(SNT) and RT-PCR,BamH I,Hind III digestion and recombinant plasmid amplified all gained the expected amplification segment.The result of sequencing showed that the PCR product shared 82.0%~94.8% homology with the NADL sequence of BVDV searched in gene bank.【Conclusion】It was confirmed that the eight isolated strains were BVDV,named B-S-1,B-S-2,B-S-3,B-S-4,B-S-5,B-S-6,B-G-1 and B-G-2 respectively.
