The Effect of Alkaloid Achyranthes Aspera Linn On Cytochrom C and APAF 1 Expresion on Mice Breast Cancer Was Induction By Benzopyrine

2017
In vivo the alkaloid Achyranthes aspera Linn to cause apoptotic and used therapeuticson breast cancer was induction by benzopyrine. The purpose of this research was provethe effect of alkaloid Achyranthes aspera Linn on mechanism of apoptotic on mice breastcancer inductions by benzopyrine on apoptosome forms on mitochondria, as cytochromC and apoptotic protease activating factor-1 (apaf-1) to induce caspase 9 dan caspase 3caused of chromosome fragmentation. This research was conducted using 50 mice femalebreast cancer induction by benzopyrine and 10 female healthy mice as negative groupscontrol , in three month of age and devided six acak groups, such as negative control, positive control was treatment by methotexat 15 mg/kg.bw, and first, second, third andfourth treatment groups each were given alkaloid Achyranthes aspera linn in dose 0 mg/kg bw, 30 mg/kg.bw, 60 mg/kg.bw and 100 mg/kg bw, All of groups was treatment forin 8 weeks. The parameter of this research showed cytochrom C dan apaf-1 expresion inbreast cancer. The alkaloid Achyranthes aspera linn in dose 60 mg/kgbb to cause apoptoticwith expresion of Cytochrom C are 106,4±4,76 and APAF-1 are 111,9 ± 9,26. Thisresult will be used mechanism of apoptotic on mice breast cancer on apoptosome formson mitochondria, as cytochrom C and apoptotic protease activating factor-1 (APAF-1).Conclution, The alkaloid Achyranthes aspera linn caused cells death of breast cancer byapoptotic mechanism to increased cytochrom C and APAF1 expresion.
