Gene coding for poly-kringle plasminogen activator and vector containing the same

PURPOSE: To obtain a gene having a base sequence coding for a hybrid plasminogen activator(PA) containing a heterologous polypeptide kringle, providing a PA having high stability and bond affinity for fibrin. CONSTITUTION: A DNA coding for urokinase is treated with a restriction enzyme so that a urokinase kringle sequence containing one kringle region composed of a nucleotide coding for a protein corresponding to amino acid 1-131 of urokinase is bound to a DNA unit containing two kringle regions composed of a nucleotide encoding a polypeptide corresponding to amino acid 92-527 of a tissue plasminogen activator (t-PA) to give the objective gene containing a nucleotide sequence coding for a hybrid plasminogen activator of a urokinase type plasminogen activator (u-PA) containing a heterologous polypeptide kringle and t-PA.
