Preparation of meniscal implant tissue

2007
A method of preparing meniscal tissue for subsequent implantation or transplantation into a host comprises freeze/thawing the tissue, incubating the tissue in a hypotonic solution (preferably 10mM Tris with EDTA and aprotinin), incubating the tissue in a hypotonic solution comprising an anionic detergent such as sodium dodecyl sulphate (SDS), repeating these incubation steps, incubating the tissue with at least one nuclease (preferably RNase and DNase), and washing the tissue with an oxidising agent such as peracetic acid (PAA). The meniscal tissue may first be ultrasonicated. The method results in a substantially decellularised meniscal tissue that may be used to repair a damaged knee meniscus.
