Characteristics of gut microbiota in southern Chinese population with colorectal adenoma based on 16S ribosomal DNA

Objective To investigate the structure and diversity of gut microbiota in southern Chinese population with colorectal adenoma. Methods The metagenomic DNA of the samples was extracted, and the V4 variable region was amplified. Using the 16S ribosomal DNA (rDNA) high-throughput sequencing and bioinformatics, we analyzed the diversity of adenoma and para-adenoma mucosa-associated gut microbiota in 36 healthy volunteers and 49 patients with colorectal adenoma. The distribution and biological markers of the phyla, families, genuses and species of gut microbiota were also analyzed. Results The diversity and distribution of mucosa-associated gut microbiota in healthy control group and colorectal adenoma group were similar (P>0.05) . The diversity of mucosa-associated gut microbiota in the colorectal adenoma group significantly decreased (P<0.05) . The distribution analysis of the gut microbiota showed significant reduction in the abundance of Firmicutes, Bacteroidetes, Lachnospira, Blautia, Coprococcus, Bacteroides-ovatus (P<0.05) , and increase in the abundance of Proteobacteria phylum, Halomonadaceae and Shewanella-algae (P<0.05) . Conclusion High-throughput sequencing confirms that there are specific diversity and characteristics of gut microbiota in patients with colorectal adenoma, suggesting that the change in characteristics of the gut microbiota may be involved in the occurrence of colorectal adenoma.
