A Method of Raising the Detecting Sensitivity to Plasmid DNA of Gel Electrophoresis

Influence of exciting wavelength on DNAEB fluorescence intensity was compared in detecting plasmid DNA of gel electrophoresis. It was found that short exciting wavelength enhanced the detecting sensitivity to DNA fractions. Using 260 nm as the exciting wavelength, as little as 0.7 ng linear DNA was measurable, and the fluorescence intensity of DNAEB complex proportioned to DNA quantity of a wide range. Furthermore, measuring irradiationinduced DNA strand breaks with this modified method, G(SSB) and G(DSB) consisted with other researches were obtained.
