Massively parallel functional dissection of mammalian enhancers in vivo

Enhancers in mammalian genomes can be hundreds or thousands of bases long, making it difficult to systematically dissect the effects of mutation on each nucleotide. Patwardhan et al. assemble short oligonucleotides into thousands of enhancer variants up to ~600 nt long and analyze their function in vivo in mice.
