RP-HPLC determination of aristolochic acid I in rat plasma

Objective:To establish an RP-HPLC method for the determination of aristolochic acid I in rat plas- ma.Metbods:Kromasil C_(18)column(250 mm×4.6 mm,5μm)was used and the mobile phase consisted of metha- nol-water-glacial acetic acid(69:30:1)at the flow rate of 1.0 mL·min~(-1).The UV-detection wave length was 250 nm and the column temperature was 35℃.Results:The linear range was 0.02-40.0 mg·L~(-1),and the limit of quantitation was 0.02 mg·L~(-1).The intra-day and inter-day RSD were less than 1.0%,and the average re- covery was between 82.4%-101.2%.Conclusion:The method was sensitive,specific and suitable for the analysis of concentration of aristolochic acid I in rat plasma.
