Production of Recombinant Human Thyroid Peroxidase in Chinese Hamster Ovary Cells

We developed a serum-free or weak serum-containing cell culture system using UC203 (product name; UC212) medium to get recombinant hTPO (rhTPO) from chinese hamster ovary (CHO) cells transfected with the cDNA for human thyroid peroxidase (hTPO), and the purification procedure of rhTPO. As a result, we could achieve the high yield and the low cost of the production by using the culture system. Furthermore, the purified rhTPO showed the similar immunoreactivity to native hTPO, which suggested that EIA using rhTPO suited for the clinical usage of testing of antiTPO antibodies in patient sera with autoimmune thyroid diseases.
