Screening of specific Z chromosome molecular markers in silkworm sex-linked balanced lethal strain through AFLP.

Silkworm Ping 48 strain(P48 in short) was obtained by transforming of group sex ratio-controlled letal genes into normal strain Baiyun,which was a near isogenic line of P48 strain.P48 and Baiyun strains were analyzed by amplification fragment length polymorphism (AFLP) in present paper for screening the Z chromosome markers in silkworm sex-linked balanced lethal strain.C094,one Z linked marker,was found by DNA retrieving,cloning,sequencing,and blasting silkworm (P50) whole genome database on NCBI.PCR primers were designed to perform the difference identification among P48,P50 and their F1 hybridization.The results showed that C094 marker was genetically linked with l1 lethal gene in silkworm balanced sex-linked lethal strain.This benefits the characterizing and cloning of lethal related genes in silkworm sex-linked balanced lethal strain.
