The present invention discloses a preparation method of fermentation broth extract having chymosin activity, comprising the steps of: using a wheat bran medium for Paenibacillus CGMCC? No.8333,15 ℃ ~ 40 ℃, shaking cultured for 18 to 120 hours to give the fermentation broth, and the resulting supernatant was centrifuged after fermentation to obtain. The resulting extract chymosin activity up to 6469.72 ± 280.65SU / mL, and its proteolytic activity only 10.54 ± 0.65U / mL, the ratio between them of up to 613.82. Using this broth extract liquid milk curd can occur in a short time, and low non-specific hydrolysis of the casein. The present invention discloses a high yield of fermentation broth extracts prepared cheese curd product having the enzymatic activity, the fermentation broth extract has broad application prospects in the original cheese made in production and processing industry.
