Стерилизация многослойных протеолипосом

The aim of this work was to investigate the possibilities and limitations of gamma-irradiation as a sterilization method for freeze-dried multilayer proteoliposomes. Freeze-dried liposomes with cell lines lysate mel P, mel Kor, mel Mtp, mel Is, mel Z were irradiated by 60Co source with doses of 5; 12; 17 and 25 kGy. The optimal dose of у -irradiation was 12 kGy as while ensuring sterility preserved basic physical and chemical properties of multilayer proteoliposomes. No changes in liposomal shape, size and structure were registered after у -irradiation with dose of 12 kGy as demonstrated by optical and electron microscopy. Immunoblot method showed that after y-irradiation of freeze-dried liposomes stability of the protein hsp-70 and a high degree of loading of the lysate into liposomes was preserved. The results obtained provide the basis for application of y-irradiation sterilization of liposomal vaccines.
