Studies on aqueous polymer two-phase systems containing agarose

Abstract Methods for high temperature preparation and separation of aqueous polymer two-phase systems containing agarose have been developed. A number of water-soluble polymers have been screened for polymer incompatibility and two-phase formation together with agarose. A method for analysis of the polymer phase composition based on chromatography at 70°C on Superose 12®, a highly cross-linked, 10 μm diameter agarose gel filtration medium, is presented. Phase diagrams for three agarose-poly(ethylene glycol) (PEG) and three agarosedextran two-phase systems are described.
