Development and preliminary application of multi-PCR in differentiation Brucella spp.

A multi-PCR assay was developed to differentiate Brucella spp.on basis of BCSP31 gene and IS711 copy numbers.Eleven strains of Brucella (B.abortus:A19,A544,A387;B.melitensis:M111,M28,M16;B.canis:RM6/66;B.suis:S2,rS2,S1330;B.ovis:63/290) could be differentiated by this assay successfully.Sizes of 494,223,178 bp could all be amplified from strains of B.abortu,and 733,223,178 bp from B.melitensis,and 223,178 bp from B.canis,and 976,223,178 bp from B.ovis,and 285,223,178 bp from B.suis.No specific bands could be amplified from control bacteria such as E.coli,Actinobacillu spleuropneumoniae,Pasteurella multocida and Salmonella.The result showed that multi-PCR assay was specific and could be used to differentiate Brucella spp.from B.abortus,B.melitensis,B.ovis,B.suis and B.ovis.This assay provided a rapid way to investigation and identification for brucellosis.
