Linum flavumL. as host for producing 5MPT were preserved in liquid nitrogen after slow prefreezing in a deep freezer (-80°C). The development of an optimal procedure from long-term storage was investigated by the addition of various concentrations of cryoprotectant mixture and osmoticum in preculture media before and after cooling. The cryoprotectant comprising 20% sucrose and 10% DMSO and an apply of 10% sucrose before reculture provided the best result. The viability with this optimized condition was about 65% after cryocell-banking for 6 months. The level of 5MPT in recovered cells from cryopreservation was also kept stable and the production level was similar to the observed in noncryopreserved cultures
