The invention discloses a pair of Talen recognition sequences of a targeting Forkhead box n1 gene of zebra fish and a preparation method of an mRNA (messenger ribonucleic acid) for targeting knockout of the Forkhead box n1 gene of the zebra fish. Gene sequences of the pair of Talen recognition sequences are represented as SEQ ID NO.1 and SEQ ID NO.2. The preparation method of the mRNA comprises the following steps: (a), the Talen recognition sequence pair in claim 1 is assembled and synthesized; (b), an expression vector of a gene knockout effector protein Talen of the targeting Forkhead box n1 gene is constructed according to the Talen recognition sequence pair in the step (a); (c), the vector obtained in the step (b) is subjected to lipofection transfection, and the protein expression condition is detected in a 293T cell; and (d), the vector obtained in the step (b) is subjected to in vitro transcription, and the mRNA for targeting knockout of the Forkhead box n1 gene of the zebra fish is obtained. The obtained mRNA can be used for knocking out the Forkhead box n1 gene of the zebra fish, and a zebra fish thymus development study model is prepared. Compared with the prior art, the cost is low, the design is simple, the success rate is high, and an obvious effect is realized.
