Encapsulation of oxaliplatin in nanostructured lipid carriers- preparation, physicochemical characterization and in vitro evalulation

Many drugs with high lipophilic character incorporated into nanostructured lipid carriers (NLC) have been reported. However, hydrophilic drugs have not been completely investigated. In this paper, we investigated the potential of lipid nanoparticles as carriers for hydrophilic oxaliplatin. NLC was prepared by hot high-pressure homogenization method which can be used in the industrial production conveniently and characterized by dynamic light scattering and transmission electron microscopy for size and morphology. Differential scanning calorimetry and X-ray diffraction studies revealed that the drug was dissolved in the blend of glycerol monostearate and medium chain triglycerides with amorphous forms. Encapsulation efficiency was determined by high performance liquid chromatographic method using an ultrafilter. The in vitro release experiments of NLC exhibited biphasic drug release pattern with burst release at the initial 2 h and prolonged release afterwards.
