Biocompatibility of Ti Coatings with Varied Thicknesses on Ti Alloy Substrates using Osteoblasts

The purpose of this study is to investigate cellular biocompatibility of Ti coatings with varied thickness on titanium alloy substrates using osteoblasts and to explore the surface characteristics of implants, which are in favour of the growth of osteoblasts. Cellular behavior is closely related to the intersurface properties between host and implant. Ti is a type of biocompatible biomaterial. Sandblasting on titanium alloy will form a metal protective layer, which will prevent the release of toxic metal ions from titanium alloy. At the same time, it will increase the surface roughness as well as the area for cell growth. The intersurface roughness of Ti coatings with varied thicknesses is quite different. By comparing the growth and proliferation of osteoblasts grown on samples with varied thicknesses, a series of cell testings were performed. The proliferation ability of osteoblasts was measured by MTT assay, the cell activity was studied by CCK-8 assay, the surface of Ti coatings was characterized by scanning OM (Optical Microscope). Cell morphology grown on Ti interface was observed by fluorescence staining. The effect of Ti coating on the growth and proliferation of osteoblasts was studied on the basis of surface roughness. The result shows that with the increase of surface roughness, the ability of growth and proliferation of Murine osteoblastic MC3T3-E1 increased and that the three-dimensional cell growth was developed by the large surface roughness of Ti coating, which is promote the growth and adhesion of osteoblasts. The research results can be used in bone replacement and dental applications to improve the integration of implants and human tissues.
