Pharmacophore model of growth hormone secretagogues

Growth hormone releasing peptide (GHRP) is the artificial exogenous peptide hormone that stimulates growth hormone secretion. GHRPs and their peptidomirnetics are currently being evaluated clinically as alternatives to GH replacement therapy in the treatment of growth hormone deficient children [1]. In this paper, we selected 105 non-peptide GH secretagogues [2-13] and established a pharmacophore model using distance-comparisons (DISCO) and comparative molecular field analysis (CoMFA) [14, 15]. These compounds are analogues of L-692, 429, MK-0677 and (G-7502 (fig. 1). All modeling was performed with the program Syby16.3, run on Silicon Graphics workstation.
