This method is characterized in that: (1) sequence is hybridized on which the base is located to identify a sufficient length nucleotide; (2) is started the synthesizing the complementary strand of the hybrid obtained in 1, - said nucleotide serving as a primer -, in the presence: - of a polymerase without 3'5 'exonuclease action' and - at least one modified nucleotide base; (3) detecting the blocking nucleotide base incorporated by any suitable means .
