Synthesis and Application of a PEGA Polymeric Support for High Capacity Continuous Flow Solid-Phase Peptide Synthesis.

Abstract Bisacrylamido polyethyleneglycol, N , N ′-dimethyl acrylamide and acrylol sarcosin ethyl ester were copolymerize by inverse suspension polymerization to obtain a beaded solid support. The resin was trans-amidated with ethylenediamine and functionalised to high capacity with 4-hydroxymethyl benzamido anchoring group. The application of newly developed resin is illustrated by the synthesis of two peptides in up to 89% overall yield and high purity. The pure peptides were characterized by laser-desorption mass spectrometry and amino acid analysis.
