The effect and mechanism of extract ginkgo biloba(EGb) on learning and memory ability of Alzheimer's disease model of rats injected β-amyloid protein(Aβ)

Objective: To investigate the effect and mechanism of action of extract ginkgo biloba(EGb) on learning and memory ability of rats injected β-amyloid protein.Methods: Y-electric maze was used to detect learning and memory ability of Alzheimer's disease model of rats injected β-amyloid protein (Aβ),with hematoxylin-eosin (HE) staining,TUNEL method and immunohistochemistry staining to detect changes of hippocampal neuronsmorphology,apoptosis of hippocampal neurons ,expression of Caspase-3P20 and deposit of Aβ in the CA1 subfield ,and to observe the effect of EGB.Results: The model rats injected Aβ1-40 showed their disorders of learning and memory abilities detected by Yelectric maze;neurons in hippocampal CA1 subfield exhibited destroyed-former lines,TUNEL positive cells and Caspase-3P20 immunoreactive positive cells and there were Aβimmunoreactive deposits .EGb-administered groups were improved.Conclusion: Aβ induced neuronal apoptosis of hippocampal CA1 subfield in rats injected Aβ into this region concerned with activation of Caspase-3.EGb attenuated this experimental AD rat's learning and memory impairment and had protective effect.
