Preparation and characterization of the monoclonal antibodies against nucleocapsid protein of IBV.

Monoclonal antibody(MAb) hybridoma cell lines 2D2 that secreted MAb against N protein of avian infectious bronchitis virus were developed by fusion myeloma cells with spleen cells of BALB/c mouse immunized with IBV CK/CH/LDL97 F115 virus.The MAb 2D2 belonged to IgG1 subtype with κ chain.ELISA and western blot assays showed that MAb 2D2 reacted specifically with N protein of IBV.The MAb 2D2 can also react with N protein of several different IBV serotypes,indicating that the epitope of the 2D2 may locate in the conserved region of IBV.This MAb provided a foundation for study of genetic variation of IBV and for the development of diagnostic reagent.
