Enhancement of gene expression of interstitial collagenase in rat hepatic stellate cells by recombinant human augmentor of liver regeneration

Objective To investigate the effects of recombinant human augmentor of liver regeneration (hALR) on interstitial collagenase(MMP 13) gene expression in hepatic stellate cells.Methods Hepatic stellate cells (HSC-T6) were cultured in medium containing different concentrations of hALR (200,20,2 μg/L respectively) and cells were collected at different incubation periods (8,24,48,72 h).Total RNA of HSC was isolated and MMP 13 gene expressions level was detected by reverse transcription polymerase chain reaction.Results MMP 13 gene expression levels of HSC in high (200 μg/L),mid (20 μg/L) and low (2 μg/L) concentrations of three hALR groups were much higher than those in the control group after exposure to hALR at 8,24,48 and 72 h.The maximum values appeared at 24 to 48 h.The expression level in low dose group,mid dose group and high dose group was 3 times,4 times and 6 times of control group,respectively.Conclusion hALR could promote gene expression of interstitial collagenase in hepatic stellate cells.
