Comparisons of Different RNA Interference Vectors for Primary Liver Cancer Cell Lines HCCLM6

Objective:To screen out the efficient and relatively safe RNA interference vector for HCCLM6 cell. Methods: HCCLM6 cells were transfected with the non-specific short hairpin RNA (shRNA) by lentiviruses, liposomes or magnetic nanoparticles.Transfection efficiencies of different vectors were detected by flow cytometer. Change of cell growth curve was measured by MTT method. Results:Transfection efficiency of lentiviruses was highest (90%) followed by liposomes (60%). Magnetic nanoparticles were the least efficient vector (30%). Compared to the wild type strain, cells transfected by lentiviruses and liposomes grew slowlier without statistically significance (P0.05). Little variance was found between cells transfeced with magnetic nanoparticles and wild type strains (P0.05). Conclusion: Lentiviruses were both efficient and relatively safe vector for HCCLM6 cell lines.Transfection methods of magnetic nanoparticles need further optimization.
