Determination of rosmarinic acid in seeds and oils of Perilla frutescens(L.)Britton

The contents of rosmarinic acid(RA)in seeds and oils of Perilla frutescens(L.)Britton were determined by RP-HPLC.Rosmarinic acid was separated and analyzed on Kromasil C18 column(250mm×4.6mm,5μm)using methanol-phosphoric acid solution(40:60)as mobile phase at flow rate of 0.8mL/min.UV detection wavelength was set at 210nm and the column temperature was 28℃.An average recovery of 97.3% was obtained with a RSD of 2.1% in the range of 0.20~4.00μg for rosmarinic acid.The linear regression equation of dyclonine hydrochloride was Y=3.02×105X-3.50×103(r=0.9999)for HPLC method.The contents of rosmarinic acid in seeds and oils were 0.2475mg/g and 0.0794mg/g.The content of rosmarinic acid in seeds was 3.12times higher than that of in oils.Active Component of rosmarinic acid remained mostly in oil meal by mechanical extrusion.The method was sensitive and accurate.It has good repeatability and a proper method for determination of rosmarinic acid in seeds and oils of Perilla frutescens(L.)Britton.
