Correlation of carcinoembryonic antigen-related cell adhesion molecule 1 and adiponectin signaling pathway related molecules in non-traumatic necrosis of the femoral head

Objective
To detect the correlation between the expression of carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1) and adiponectin, adiponectin receptor 1 (AdipoR1), AdipoR2, nitric oxide (NO), endothelial nitric oxide synthase (eNOS) in the patients with non-traumatic osteonecrosis of femoral head (ONFH).
Methods
The serum levels of CEACAM1, adiponectin and NO were determined by enzyme-linked immunosorbent assay (ELISA) in patients with ONFH (ONFH group) and healthy control group (60 cases). The expression levels of CEACAM1, adiponectin, AdipoR1, AdipoR2 and eNOS were detected by reverse transcriptase-polymerase chain reaction (RT-PCR).
Results
The levels of serum CEACAM1, adiponectin and NO in patients with ONFH were significantly different from those in healthy controls [(5.35±2.21) ng/ml vs. (21.57±5.38) ng/ml; (4.26±1.05) μg/ml vs. (12.21±4.89) μg/ml; (10.31±2.15) μmol/L vs. (35.13±5.07) μmol/L, P<0.05]. There was significant difference in the expression of CEACAM1, adiponectin, AdipoR1, AdipoR2 and eNOS between the two groups (4.03±1.31 vs. 7.35±1.05; 10.26±2.57 vs. 20.13±3.54; 11.23±1.52 vs. 19.28±2.37; 10.19±1.18 vs. 21.72±2.19; 15.19±2.01 vs. 31.12±3.18, P<0.05); and the expression of CEACAM1 was negatively correlated with ONFH (r=-0.236, P<0.01). The levels of serum CEACAM1 in patients with ONFH were positively correlated with the levels of adiponectin and NO (r=0.179, 0.237, P<0.01). The mRNA expression of CEACAM1 was positively correlated with the mRNA expression of adiponectin, AdipoR1, AdipoR2 and eNOS (r=0.157, 0.372, 0.253 and 0.154, P<0.01).
Conclusion
The expression of CEACAM1 in ONFH patients is closely related to the expression of adiponectin signaling pathway, which provides a new way for the clinical diagnosis and treatment of ONFH.
