Application of MLPA in the genetics detection of spontaneous abortion villus

Objective Use of the multiplex ligation probe amplification(MLPA)technology to detect various chromosomal abnormalities in abortive embryos, assist patients in analysing the causes of abortion, and further counsel for eugenics. Methods The villus tissue samples of early pregnancy abortive embryos were collected and genomic DNA was extracted. MLPA-P036 and MLPA-P070 probes were used for detection. Simultaneously, gene chip techniques were used for detection. The accuracy of the two methods was compared. Results The detection rate of chromosomal abnormality in 212 abortive embryos was 50%, which could occur on many chromosomes. Among them, chromosome aneuploidy accounted for 48.58%. Most of them were sex chromosome trisomy, 16-chromosomes trisomy and X-chromosome monomer. MLPA technology can accurately detect a variety of chromosomal abnormalities including chromosome trisomy and chimerism, along with one-arm copy number abnormalities. Conclusion MLPA technology has many advantages such as accuracy, simplicity, rapidity, practicality, and low cost in the detection of chromosomes in spontaneous abortion embryos. It has significant application value in the diagnosis of abortion etiology and is worthy of clinical promotion. Key words: Spontaneous abortion; MLPA; Chromosomal abnormalities
