Modelling on the Kinetics Mechanism of the FokI Restriction Endonuclease

Fokl is a special enzyme in the family of restriction endonucleases. The Fokl restriction endonuclease recognizes an asymmetric DNA sequence and cuts both strands at fixed positions downstream of the site. Because of the high efficiency of catalyzing and the fast rate of the reaction, it is widely used in many operations in molecular biology including DNA computer. Researchers have made great effort to unveil its mechanism since it was first discovered. In this paper, the research in the reaction mechanism of Fokl is summarized, and two rational mechanisms are concluded. Then the math model based on the kinetics is set up.
