Effect of Tributyltin Exposure on Activities of Alkaline Phosphatase in Sebastiscus marmoratus

2005
The effects of tributyltin (TBT) on the activities of alkaline phosphatase (AKP) of liver, spleen and kidney in Sebastiscus marmoratus were studied at injected intraperitoneally with single doses of 1.9,9.6,19.3,193 μg /kg.After the injection 2,4 and 7 days, six fishes per dose group were killed and the liver, spleen and kidney were frozen in liquid N_2 immediately after collection and stored at -80℃ until analyzed. Each liver was measured solely, while 2 spleens or kidneys were got into a group for measure of enzymes activity,AKP activity was assayed using p-nitrophenylphosphate (pNPP) as the substrate. The results showed that the AKP activities were primarily induced in the fish exposed to TBT at low concentrations. The dose-response curve was a bell shape.AKP is participated in phosphoric metabolism directly, and participated in the metabolism of DNA,RNA,protein, lipid and so on.The results suggested that metabolism in the fish would be interfered by TBT exposure. The spleen and kidney are the immune and hemopoietic organs in fish.Effect of TBT on the activity of AKP in the spleen and kidney would affect the immune function in the fish.
