Effects of Trichostatin A on Cell Cycle Arrest and Apoptosis in Molt-4 Cells

Objective: To explore the effects of trichostatin A(TSA) on cell cycle and apoptosis in Molt-4 cells and the anti-tumoral actions.Methods: The effects of TSA on cell cycle arrest and apoptosis in Molt-4 cells were examined using cell culture,PI staining,flow cytometry and DNA Ladder.Results: Within a certain range of dose,TSA induced S phase arrest and G_2 phase arrest in Molt-4 cells in a dose-dependent manner.The apoptosis induction effect of TSA in Molt-4 cells was also dose-dependent.The apoptosis percentage increased according,with the increase of the concentration of TSA.The apoptosis percentages of the experiment groups had significant difference from the control group(P0.01).The apoptosis induction effects of TSA in Molt-4 cells were time-dependent under the treatment of 200 nmol/L TSA.Besides,TSA showing significant apoptotic cytotoxicity in Molt-4 cells did not demonstrate apparent apoptosis induction in normal human peripheral blood mononuclear cells(NPBMNC) with the definite dose range.Conclusion: TSA shows effects of definite and significant cell cycle arrest and apoptosis induction on Molt-4 cells in time-and dose-dependent manners.TSA is able to selectively induce apoptosis in Molt-4 cells with low toxicity in NPBMNC at the same time.
