Synthesis of 18F-SAN as peripheral benzodiazepine receptor tracer.

Objective:To synthesize the 18F labeled compound of N-(2,5-dimethoxybenzyl)-3-fluoro-N-(5-fluoro-2-phenoxyphenyl) propionamide(18F-SAN) according to forerunner compound of N-(2,5-dimethoxybenzyl)-N-(5-fluoro-2-phenoxyphenyl) propanamide ligands.Methods:The 18F ion and 18F-SAN precursor,3-bromo-N-(2,5-dimethoxybenzyl)-N-(5-fluoro-2-phenoxyphenyl) propanamide carried out nucleophilic substitution reaction.The labeled compounds were isolated and purified by HPLC.Results:The preliminary results showed that the radiochemical purity of 18F-SAN was more than 95%,radiochemical yield was about 50% and the total synthetic time was approximately 60 minutes.The Rf value of 18F-SAN detected by TLC was 0.4~0.5 and the tR value of 18F-SAN detected by HPLC was 6.24 minutes.
