Extraction and Analysis of DNA from Endosperm in the Seed of Pinus

Improved SDS method was effective to extract DNA from little plant seeds. Using this method, DNA of high purity was successfully obtained from the endosperm of the seeds of Pinus massoniana, Pinus taiwanensis and Pinus thunbergii. Its purity, concentration and yield were characterized. The result of RAPD examination shows that these isolated DNA(s) have good expanding effects and can meet directly the needs of PCR reaction, RAPD and AFLP analysis. Thus a rapid, economic and reliable DNA extraction procedure was developed for Pinus and the other little seed plants.
